Ben Goult (University of Kent) and I found that we both had initial data suggesting that the intracellular proteins working with integrin extracellular matrix receptors form a functional aggregate after they are unfolded by mechanical forces. This is one of a growing number of examples where protein aggregation is used in a positive way, for example as seen in the process of memory formation, and contrasts with the deterimental aggregates linked to dementia. The postdoc in my lab will test the importance of self-interactions in integrin function within the fly, to complement biochemical and structural work on the formation and structure of the aggregate's performed by the postdoc in Ben's lab. The project involves extensive gene-editing followed by phenotypic analysis by advanced microscopy, so experience in molecular biology, fly genetics and development and microscopy will all be useful.
